Overview: Microbial IDentification Inc (MIDI) is a privately held, veteran-owned biotechnology company based in Newark, Delaware USA. The company has been developing rapid microbial identification solutions for clinical and environmental microbiology laboratories under the Sherlock brand name since 1991. The fatty acid-based technology behind the Sherlock brand was created in the early 1980s out of a partnership between the Hewlett-Packard Co. (later became Agilent Technologies in 1999) and Dr. Myron Sasser in University of Delaware's Plant Pathology Department. The goal of the partnership was to create a rapid phenotypic method for the identification of plant pathogenic bacteria based on gas chromatographic analysis of cellular fatty acids (GC-FAME). The original product was named the HP Microbial Identification System and released in 1985.

Fatty acid methyl ester analysis by gas chromatography (GC-FAME) has been used for over 50 years to identify microbes in environmental and clinical settings. FAME identification is a dynamic test of the organism. It is specific to growth conditions, including the media and temperature used. Different libraries are available for different growth conditions and custom libraries can be created based on the customers unique environment. In addition to identification, FAME analysis is often sensitive enough to give strain level information for a sample.

Sherlock Mycobacterial ID System (MYCO-LCS)
Mycolic acid analysis by High Performance Liquid Chromatography (HPLC) has been used for more than 15 years by mycobacteriology laboratories worldwide to aid in the diagnosis of tuberculosis and other mycobacterial infections. The technology is much less expensive than DNA sequencing, requires considerably less labor and time than conventional biochemical identification and is not subject to erroneous identification of Mycobacterium celatum as M. tuberculosis, as may happen with DNA probes.
