The DNA sample undergoes PCR amplification using primers that flank the specific target region of an HLA locus. This is followed by hybridisation of the resulting amplicon to an array of Sequence Specific Oligonucleotide probes immobilised on a nylon membrane. The SSO probes are designed to hybridise to polymorphic target sequences in the HLA locus. The Biotest ELPHA (Enzyme Linked Probe Hybridization Assay) System is a DNA-hybridization test for the determination of HLA Class I and Class II alleles. Sequence-specific oligonucleotide (SSO-) probes are used as diagnostic tools to identify polymorphic sequence motifs. The hybridization between probe and target DNA is detected by a method adapted from the protein-ELISA technique.
